Overexpression of Klotho decreases endogenous β-catenin level and inhibits its nuclear translocation. The liver cancer cells (2 × 103 cells/per well) were plated into 6-well plate, and the cells were transfected with pCMV6-Klotho and the control plasmid after 8 h. Cell lysates were prepared for detecting of β-catenin in cytoplasm and nucleus by western blot analysis after transfection for 48 h. And, the expression levels of cyclin D1 and C-myc were also detected by western blotting analysis. β-tublin was used as an internal reference in cytoplasm, and lamin B1 was used as the internal reference in nucleus
