Amiloride down-regulated the mRNA expression of urokinase (u-PA) in MKN45 gastric cancer cells. The MKN45 cells were added at the amount of 2 × 10⁶ cells per dish. After incubation for 6 h, 12 h or 24 h, cells were treated with amiloride in concentrations of 0.01 mM , 0.1 mM or 1 mM, and RPMI 1640 with 10% FBS was used as a control. Total RNA was isolated from MKN45 cells using TRIzol reagent (Qiagen, Japan) according to the manufacturer’s instructions.
